Summary

Cell surface carbohydrates play key roles in cell recognition mechanisms. Protein-carbohydrate interactions typically exhibit high specificity and weak affinities toward their carbohydrate ligand. This low affinity is compensated in nature by the architecture of the protein, the host presenting the carbohydrate ligands in a multivalent manner or as clusters on the cell or mucosal surface. O-glycosylation is a ubiquitous post-translational modification that is highly dynamic and responsive to cellular stimuli through the action of the cycling enzymes. Expression of specific O-glycans is linked to changes in gene expression in, for example, inflammatory bowel disease, cystic fibrosis and several types of cancer. Understanding these glycosylation patterns at molecular and functional levels will allow mechanisms associated with bacterial-host interactions, bowel disease and other cancers to be defined, which will facilitate the development of new effective therapeutics and diagnostic tools for these Conditions.
In this project, the chemical synthesis of novel multivalent mucin-type O-glycan probes for the screening of O-glycosylation-linked interactions in health and in disease and the development of smart glyco-nanoparticles as drug delivery systems have been addressed.

synthesis of novel functional smart glyco-nanomaterials that can target diseased cells over healthy cells and the fundamental understanding of glycan mediated processes is our main objective in this proposal. Its potential impact upon society (novel diagnostic tools and therapeutics – personalised medicine) means that aspects of the project will be of high impact and of general interest and for society.

Work performed

The proposed work delivered novel glycan tools that have allowed us to study the mechanisms associated with diseases like cancer, also we prepared fucose derivatised glycans to study bacterial glycan towards antibiotic preparation. The knowledge from these glycan interactions has been used to developing a new class of targeted glycan-based drug delivery systems that are tailored to a specific disease. Thus, this programme has shown great use of glycans in the fields of chemistry, glycobiology and glycomedicine.

Final results

Harnessing chemical technologies effectively, sustainably for drug development for life-threatening diseases are the key element for achieving EU sustainable development. This project introduces an efficient methodology for the development of a targeted delivery system using glycan derivatives. This will contribute to ensuring the EU sustainable future. The glycan-based drug delivery systems detailed in this action has tremendously impacted academia, triggering growing interest in the topics discovered.
